RHEOL® SNAC

RHEOL® SNAC, chemically known as 8-(2-hydroxybenzamido)octanoic acid sodium salt, is a white to off-white crystalline solid with good water solubility. It is a high-performance oral permeation enhancer that locally elevates gastric pH, promotes peptide monomerization, and anchors absorption in the stomach to significantly improve oral bioavailability.

Product Introduction

RHEOL® SNAC is one of the most clinically validated oral absorption enhancers in the pharmaceutical industry, designed as a pharmaceutical excipient that adheres to the gastric mucosa alongside peptide actives. It creates a localized microenvironment that protects the drug from peptic degradation, drives peptide monomerization for enhanced permeability, inserts into the gastric epithelial cell membrane to enable transcellular transport, and anchors the absorption site in the stomach to maximize oral bioavailability. RHEOL® SNAC delivered by Westpoint offers formulation scientists a reliable enabling excipient for the next generation of oral peptide and macromolecule therapeutics.

Product Feature

  • Gastric-targeted absorption
  • Local pH elevation
  • Peptide monomerization
  • Membrane insertion & transcellular transport
  • Enhanced oral bioavailability
  • Good water solubility
  • Crystal form consistent with originator

Product Application

  • GLP-1 Receptor Agonist (Semaglutide)

The oral semaglutide (Rybelsus®) developed by Novo Nordisk is co-formulated with 300 mg of SNAC and 7 mg or 14 mg of semaglutide. Validated by the PIONEER 10 phase III clinical trials, its glucose-lowering and weight-loss effects are comparable to subcutaneous injection.

  • Abiraterone Acetate
NMPA approved SNAC for use in a nanocrystal formulation of abiraterone acetate (a Class 2.2 improved new drug).

  • Heparin (Anticoagulant)
After oral administration of the heparin–SNAC complex, activated partial thromboplastin time (APTT) and anti-Xa factor levels significantly increase within 15 minutes and remain elevated for over 4 hours. Compared with subcutaneous low-molecular-weight heparin, the incidence of thrombosis and thrombus weight are significantly reduced, with comparable efficacy between the two.

  • Octreotide
Ex vivo model studies show that at a concentration of 20 mmol/L, SNAC significantly increases the apparent permeability coefficient of octreotide across different rat intestinal segments: stomach (1.3→1.9×10⁻⁶ cm/s), duodenum (2.1→3.0×10⁻⁶ cm/s), and colon (2.5→7.9×10⁻⁶ cm/s), showing a trend of increase from proximal to distal.

  • Vitamin B12
The vitamin B12–SNAC co-formulation has been approved as a medical food and granted FDA GRAS status, representing one of the earliest commercialized applications of SNAC technology.
